从活性污泥中分离到1株能够利用丁酸合成聚羟基脂肪酸酯(PHA)的细菌WD-3,对该菌株积累PHA的发酵特性进行了研究,包括PHA产量随碳氮比、pH、发酵时间的变化,并考察了整个发酵过程中碳源、氮源、生物量、pH值和PHA产量之间的关系.结果表明,当C/N为35,pH为7.0时,WD-3的PHA合成能力最高.在最佳培养条件下,细胞生长达到稳定期时,该菌PHA产量高达3.01 g·L-1,占细胞干重的45.4%,且PHV所占比例达到三分之一.经16S rRNA基因测序分析,初步确定该菌属于γ-变形菌亚纲中的肠杆菌属Enterobacter. |

A bacterial strain,WD-3,with high polyhydroxyalkanoates (PHA) yield was isolated from activated sludge. Its fermentation characters including the effects of C∶N ratio,pH,fermentation time on the PHA production were also primary studied. The results showed that the optimal C/N ratio and pH were 35 and 7.0,respectively.PHA accumulation increased rapidly during the stable growth stage.The maximum PHA yield was 3.01 g·L-1,the strain accumulated PHA up to 45.4% of its cellular dry weight,and the proportion of PHV occupied one-third of the PHA production.Analysis of its 16S rRNA sequence showed that strain WD-3 belongs to Enterobacter. |
