微藻-细菌共生系统对抗生素抗性基因赋存及传播影响的研究进展

摘要: 抗生素抗性基因(ARGs)的传播已成为全球公共卫生领域的重大挑战,尤其是在污水处理厂这一关键环节. 随着抗生素的广泛使用,耐药性细菌和ARGs在环境中的传播速度加快,污水处理厂成为ARGs扩散的重要源头之一. 传统污水处理技术如活性污泥法和膜过滤法等虽然能去除污水中的部分有机污染物和微生物,但对于ARGs的去除效率较低,且常伴随高能耗和化学试剂使用,存在二次污染的风险. 因此,寻找高效、环保和可持续的污水处理技术成为当务之急. 菌藻共生系统作为一种新兴的污水处理技术,通过微藻与细菌的协同作用,展示了在去除ARGs方面的巨大潜力. 微藻通过光合作用固定二氧化碳,吸收水中的营养物质,而细菌则发挥其丰富的降解能力,能够有效降解水中的ARGs及其他有害物质. 菌藻共生系统不仅能够高效去除ARGs,而且由于其低能耗和无需大量化学试剂,符合绿色和可持续的处理要求. 微藻-细菌之间的相互作用,包括代谢协同与群体感应机制,是该系统去除ARGs的关键. 然而,菌藻共生系统在实际应用中仍面临诸多挑战,尤其在系统稳定性、规模化应用和运行成本等方面. 此外,群体感应系统在调控ARGs的传播和去除上发挥了重要作用,微藻通过调控细菌群体感应,可能增强ARGs去除效果并抑制其水平转移. 本文综述了菌藻共生系统在ARGs去除中的应用、去除机制、环境影响、面临的挑战及未来的研究方向,提出了优化该技术系统稳定性和实现规模化应用的建议,并展望了其在污水处理领域的广泛应用前景.

Research Progress on Microalgae-bacteria Symbioses Influencing the Persistence and Spread of Antibiotic Resistance Genes

Abstract: The dissemination of antibiotic resistance genes (ARGs) has become a major challenge in global public health, particularly in wastewater treatment plants, which serve as critical hubs. With the widespread use of antibiotics, the spread of resistant bacteria and ARGs in the environment has accelerated, making wastewater treatment facilities a significant source of ARG dissemination. Traditional wastewater treatment technologies, such as the activated sludge process and membrane filtration, can remove some organic pollutants and microorganisms from water, but their efficiency in eliminating ARGs remains low, often accompanied by high energy consumption and the use of chemical agents that risk secondary pollution. As a result, the development of efficient, eco-friendly, and sustainable wastewater treatment technologies has become an urgent priority. The microalgae-bacteria symbiotic system, an emerging approach, exhibits immense potential for ARG removal through the synergistic interplay of microalgae and bacteria. Microalgae fix carbon dioxide via photosynthesis and absorb nutrients from water, while bacteria utilize their robust degradative abilities to effectively break down ARGs and other harmful substances. This system not only excels in removing ARGs but also meets green and sustainable standards due to its low energy demands and minimal reliance on chemical reagents. The interactions between microalgae and bacteria, including metabolic cooperation and quorum sensing mechanisms, are central to its effectiveness in ARG removal. However, practical application of the microalgae-bacteria symbiotic system faces challenges, particularly in terms of system stability, scalability, and operational costs. Furthermore, quorum sensing plays a vital role in regulating ARG dissemination and removal, with microalgae potentially enhancing ARG elimination and inhibiting their horizontal transfer by modulating bacterial quorum sensing (QS). This review examines the application of the microalgae-bacteria symbiotic system in ARG removal, its underlying mechanisms, environmental impacts, challenges, and future research directions, offering suggestions to improve system stability and achieve large-scale implementation, while highlighting its promising prospects for widespread use in wastewater treatment.

抗生素抗性基因(antibiotic resistance genes,ARGs)的传播已成为全球公共卫生领域的重大挑战之一[1]. 近年来,随着抗生素的广泛使用和滥用,耐药性细菌的数量逐渐增加,这些耐药性细菌对传统抗生素的治疗效果大大降低,威胁着人类健康以及动物和生态环境的安全[2]. ARGs通常存在于水体和土壤等环境中[3,4]. 污水处理厂作为污水处理的重要环节,因其能够接触到来自医院、农业和工业等多个领域的抗生素污染,成为ARGs传播的重要场所. 污水处理厂不仅是ARGs的“宿主”,更是ARGs向环境中扩散的关键点[5,6]. 如果污水处理系统不能有效去除ARGs,它们可能通过污水和污泥等途径污染水体和土壤,从而促进耐药基因的传播,进一步威胁到生态环境和公共卫生.

传统的污水处理方法,如活性污泥法、化学沉淀法和膜过滤法等,长期以来一直是解决水体污染的主要手段[7,8]. 然而,以上传统技术在去除ARGs方面存在一定的局限性. 作为生化法典型代表的活性污泥工艺虽然可以有效去除污水中的有机污染物和部分微生物,但对ARGs的去除效率相对较低,甚至可能导致其富集[9,10]. 同时,ARGs在污泥相中显著积累,从而增加了后续污泥处理与处置过程中ARGs扩散的风险[11,12]. 此外,传统工艺普遍能耗高和依赖化学药剂,不仅推高运行成本,还可能带来二次污染[13~15]. 因此,开发更加高效、环境友好且可持续的新型污水处理技术已成为当前领域亟待解决的关键问题.

菌藻共生系统作为一种新兴的污水处理技术,凭借其独特的藻-菌协同作用展现出显著优势[16]. 该系统通过微藻光合作用固定CO2并去除氮磷,细菌则降解有机物和污染物,共同实现低能耗和低化学药剂依赖的高效处理与资源回收[17]. 菌藻共生系统的优势在于其能够实现低能耗和高效处理,并且不依赖于大量的化学试剂,这使其成为一种绿色和可持续的污水处理新技术. 特别是在处理ARGs污染方面,微藻-细菌协同作用可以通过直接吸附、降解以及转化等多种机制有效去除水中的ARGs,从而减少ARGs对水体和土壤等生态系统的污染风险. Cheng等[18]的中试研究表明,基于Galdieria sulphuraria的菌藻共生系统可更高效地去除ARGs,并抑制噬菌体介导的水平基因转移(horizontal gene transfer,HGT). 相比之下,在传统活性污泥工艺中,多数ARGs及intI1的相对丰度则呈上升趋势. 另一项由Cheng等[19]开展的中试研究也表明,藻菌共生系统相较于传统活性污泥系统能够更加高效去除全部类别的ARGs及多数亚型,同时通过抑制HGT和降低ARGs宿主多样性,显著减弱其环境扩散风险. 然而,目前针对菌藻共生系统消减ARGs的研究仍处于初步阶段,并多集中于宏观去除效果的观测与总结,对其中复杂的微观机制,特别是微藻与细菌间跨界胞间通讯如何介导ARGs的传播与消减等关键问题,理解仍显不足.

尽管菌藻共生系统在污水处理中的应用前景广阔,但在实践中仍然面临诸多挑战. 菌藻共生系统的运行稳定性和效率受多种因素的影响,如水质、水温和光照等条件的变化[20~22],可能导致系统的处理效果不稳定. 菌藻共生系统的规模化应用和工业化进程仍需进一步优化,需要解决微藻与细菌种群的稳定共生关系和系统运行成本等问题[23,24]. 此外,菌藻共生系统在去除ARGs的具体机制和效果仍需要进一步研究,尤其是在复杂污水中ARGs去除的具体路径和效率仍不明确.

本文旨在系统分析菌藻共生系统在ARGs去除中的应用、去除机制、环境影响、面临的挑战以及未来的研究方向. 通过对现有文献的综述,希望为菌藻共生系统在污水处理领域的广泛应用提供理论依据,并为未来相关技术的改进与发展提供思路. 在此基础上,本文还将探讨菌藻共生系统在实际应用中面临的挑战,提出相应的优化方案,并展望未来菌藻共生系统在环境治理中的发展前景.

1 菌藻共生系统在污水处理中消减耐药基因的应用 1.1 菌藻共生的生态互作基础

菌藻共生系统通过复杂的生态交互和协同代谢机制,为ARGs的去除提供了新途径. 在水处理过程中,其代谢协同效应可显著提升污染物清除效率,尤其在ARGs的消减方面展现出突出的应用潜力. 微藻和细菌之间的相互作用是菌藻共生体系稳定性和功能的基础,这种关系主要通过图 1中展示的物质交换、信息交流和基因转移这3种方式介导[25]. 菌藻共生关系在生态学上非常复杂,通常包括互利共生、拮抗作用和竞争关系等多种形式. 微藻和细菌之间的相互作用是调节菌群结构,帮助微生物群体适应外界环境变化的关键因素. 通过以上相互作用,微藻和细菌共同构成了藻-菌共生污水处理系统的结构与功能的重要调控者.

在菌藻共生体系中,物质交换是维持互利共生关系的基础. 微藻通过光合作用为细菌提供氧气和有机物,而细菌则通过呼吸作用产生二氧化碳和无机物,帮助微藻生长[13]. 此外,微藻还可以释放胞外多糖等代谢产物,如碳水化合物、氨基酸、酶、脂质、维生素和有机磷等[26,27],以上物质可供细菌利用促进其生长和代谢. 细菌能够通过合成维生素B12和吲哚乙酸等营养及信号分子,满足微藻的生理需求,进而促进其生长与代谢活性. 除了互利共生,微藻和细菌之间也存在拮抗作用[28]. 在某些情况下,微藻通过产生抗菌物质,如生物碱、脂肪酸、卤代化合物、脂质(如硫代异鼠李糖甘油二酯)以及多糖等,抑制细菌的生长,甚至杀灭细菌. 另一方面,部分细菌则通过营养竞争、释放毒素以及产生杀藻物质等方式抑制微藻的生长. 以上拮抗作用通过调节微生物群体的相对比例,进而影响系统的稳定性和功能[25].

菌藻共生系统中存在复杂的信号调控机制,其中细菌的群体感应(quorum sensing,QS)系统在维持系统协调性与功能稳定性方面发挥着核心作用. QS是指细菌通过释放并感知胞外信号分子,实现群体水平的行为调控. 在微藻-细菌的互作过程中,微藻能够释放QS信号分子类似物,从而干扰或激活细菌的QS系统,进而调节其代谢活动,如抑制细菌生长或抑制生物膜的形成. 反过来,细菌释放的QS信号分子也可影响微藻的生长状态、结构组成乃至行为表达[25,29]. 目前已识别的QS信号分子包括革兰氏阴性细菌常用的N-酰基高丝氨酸内酯(N-acyl homoserine lactones,AHLs)、革兰氏阳性细菌的自诱导肽(autoinducing peptides,AIPs)以及在不同菌种间通用的自诱导物-2(autoinducer-2,AI-2). 由于共生系统中与微藻关联的细菌多为革兰氏阴性菌,因此AHLs类信号分子被认为尤为关键. Stock等[30]研究表明N-十四烷酰基高丝氨酸内酯(C14-HSL)可促进Seminavis robusta的生长,而N-3-羟基十四烷酰基高丝氨酸内酯(OH-C14-HSL)和N-3-氧代十四烷酰基高丝氨酸内酯(oxo-C14-HSL)则表现出抑制效应. 这类基于信号分子的互作可视为化感作用的一种表现形式. 化感作用是菌藻共生系统中的关键生态调控机制,指微生物或藻类通过释放代谢产物(如QS信号分子或其类似物),对其他生物的生长、代谢或种群动态产生影响. 以上效应既包括促进,也包括抑制,涉及藻类对细菌、藻类间互作以及细菌对藻类的代谢干扰等典型类型[31]. 化感机制通过调节菌藻群落结构与功能状态,对维持共生系统的生态稳定性具有重要意义.

基因转移,尤其是HGT,是菌藻共生系统中重要的微生物进化机制,有助于细菌间遗传资源的共享与功能协同. 在特定条件下,HGT也可能发生于细菌与微藻之间,使微藻获得功能性基因,从而提升其环境适应性. 以红藻Pyropia haitanensis为例,Wang等[32]研究发现其基因组中约2.25%的基因与HGT有关,其中98%来源于细菌,主要涉及拟单胞菌门、放线菌门和拟杆菌门等类群. 核心藻类伴生细菌(如拟单胞菌门、拟杆菌门和放线菌门)可通过稳定微生物群落结构并诱导微藻中HGT来源基因的上调,从而增强其对环境胁迫的响应能力. HGT不仅增强了微藻的环境适应能力,还可能推动其与共生微生物系统的协同进化. 例如,Helliwell等[33]提出在共生细菌提供外源维生素B12的条件下,Chlamydomonas reinhardtii编码的B12非依赖型甲硫氨酸合酶基因表达受到抑制,反映出菌藻共生体系中复杂的基因调控机制. 在温度和蓝藻丰度相对较高的条件下,在大肠杆菌中含有ARGs的质粒更容易转移到蓝藻中,从而使蓝藻获得耐药基因表现出耐药性[34,35].

总的来说,微藻-细菌之间的相互作用在菌藻共生系统中起着至关重要的作用. 通过代谢产物的交换、信号分子的调控以及基因转移等机制,微藻和细菌能够在复杂的污水环境中有效协同工作,从而实现ARGs的去除. 以上生态互作关系不仅为微生物群体提供了生存优势,也为污水处理技术提供了新的思路,尤其在ARGs的去除方面展现了巨大的应用潜力.

1.2 培养方式对ARGs去除效能的影响

培养方式对微藻-细菌共生系统在去除ARGs方面的表现有着重要影响. 如图 2所示,常见的培养模式包括悬浮培养、生物膜培养和颗粒污泥培养[36]. 不同方式不仅影响微生物的空间结构与附着形式,还会调控其群落组成、代谢活性及微生物间的互作机制,从而导致ARGs去除效率与机制的显著差异.

利用悬浮培养的微藻-细菌共生系统去除废水中污染物的技术,具备操作简单和便于控制与观察的优点,且其可行性与应用潜力已得到广泛研究[13,36]. 然而,尽管悬浮态微藻-细菌共生体在废水处理领域展现出诸多优势,但其实际工程应用仍面临关键挑战. 由于微藻生物量呈分散状态,难以实现高效分离与收获,这一问题不仅直接影响出水水质的稳定性,更成为制约该技术规模化落地的核心瓶颈[37,38]. 相较于悬浮态微藻-细菌共生体,附着态微藻-细菌共生体(生物膜)展现出更优性能,包括更高的生物量产率、更低的空间占用、更好的废水净化效果、更短的水力停留时间以及更简便的生物量收获方式,以上特性为后续资源回收下游处理工序的推进创造了有利条件[39~43]. 生物膜培养通过将微生物附着在载体表面,形成稳定的微生物群落,从而提高处理过程的稳定性和耐受性. 生物膜系统特别适用于处理复杂的污水,能够提供更大的微生物接触面积,并促进微生物群体之间的相互作用[42]. 在菌藻共生系统中,微藻和细菌通过代谢产物的交换和物质传递维持着稳定的互利共生关系. 生物膜的形成不仅有助于提升微藻-细菌协同代谢的效率,还能提高对ARGs的吸附和固定能力. 生物膜的结构有助于提高系统的稳定性,并能有效地吸附和固定ARGs,从而减少ARGs在水中的迁移和扩散[44]. 颗粒污泥作为一种新型的污水处理方式,具有高密度的微生物群落,能够在较高的污水负荷下维持较高的处理效率. 颗粒污泥的结构特点使其能够在较短时间内快速沉降,便于污水的后续处理与分离[45]. 细菌在颗粒污泥中的浓度较高,这有助于增强抗性菌的降解或竞争能力,从而提高系统对ARGs的去除效率. 颗粒污泥通过高浓度的细菌群体,能更有效地降解或竞争ARGs,从而减少抗性基因的传播与扩散.

近年来微藻-细菌颗粒污泥(microalgae-bacterial granular sludge,MBGS)系统作为一种具有代表性的且新颖的方式,因其低能耗、温室气体排放少和资源回收潜力大等优点,受到了广泛关注[46,47]. MBGS体系中的颗粒污泥不仅具有优良的沉降性能,能够有效降低污水处理过程中的碳排放和能耗,还能够收获微生物油脂和生物肥料等高附加值产品,推动污水处理实现减污降碳的协同增效[48]. 据Zhang等[49]研究报道,MBGS工艺中生物质经厌氧消化回收的能量不仅能抵消系统与曝气和污泥分离相关的运行能耗,还可实现可观的净收益,每年净能量收益达641亿kW·h,同时减少6 020万t二氧化碳当量的温室气体排放. 因此,发展MBGS工艺对提高污水处理效果,尤其是对减少ARGs和耐药细菌的丰度具有重要意义. Liu等[50]的研究表明,由内源微藻和好氧颗粒污泥构成的MBGS体系,在四环素和磺胺嘧啶的去除率方面,比单独的BGS体系更高,且ARGs及其宿主的丰度明显降低. 此外,Kiki等[47]基于外源微藻Haematococcus pluvialis构建的MBGS系统,抗生素去除率较传统BGS系统提高了5.2%~8.2%,同时,MBGS出水中和生物质中的ARGs和移动遗传元件丰度较低. 尽管如此,目前关于MBGS系统在去除污水中ARGs的研究尚处于起步阶段,且大多聚焦于抗生素及ARGs的去除,忽视了对耐药细菌的监测和控制. 因此,进一步研究MBGS系统在去除污水中耐药细菌及ARGs方面的综合效果,将为其应用提供更为广泛的理论依据.

综上所述,培养方式对微藻-细菌共生系统在去除ARGs中的效果有着至关重要的影响. 不同培养方式通过调节微生物的群落结构、代谢活动以及协同降解机制,能显著改变ARGs的去除路径,如吸附与降解. 尤其是MBGS体系,在低能耗、资源回收及高效去除污染物方面的优势,展示了其作为一种新型污水处理技术在减少ARGs传播中的巨大潜力.

1.3 菌藻共生系统消减耐药基因的环境与培养条件影响因素

菌藻共生系统消减ARGs的效果受多种因素的影响,主要包括环境因素、菌种选择与系统稳定性、系统设计与操作条件以及抗性基因的类型和复杂性. 环境因素对菌藻共生系统的表现有重要影响. 水中的pH值、温度、溶解氧和氮磷浓度等水质条件显著影响微藻和细菌的生长及其代谢活动,从而直接决定ARGs的去除效果[51,52]. 过低或过高的pH值可能抑制细菌的降解功能,而温度和溶解氧浓度的变化则会影响微藻的光合作用效率及细菌的代谢活动,从而影响ARGs的去除效果. 光照条件对微藻的光合作用也具有直接影响,光照强度和时长不足时,微藻的光合作用能力会受到限制,导致微藻活性下降,进而影响整个系统的去除能力[53]. 水温的变化对微藻和细菌的代谢速率有直接影响,过高或过低的温度可能抑制微藻或细菌的活动,导致ARGs去除效果下降[53]. 此外,微藻与细菌的种群稳定性对菌藻共生系统的长期效果至关重要. 环境因素如温度、pH值和养分浓度等变化可能导致微藻与细菌之间的共生关系不稳定,从而影响系统的持续效果. 为了确保系统的高效运行,保持微藻与细菌种群的稳定性至关重要[23]. 同时,菌种多样性在系统中的应用也对ARGs去除效果有显著影响. 多样化的菌种能够增强系统对不同污水条件的适应性,并提高对不同抗性基因的降解能力[54]. 因此,选择适宜的微藻和细菌种类,并维持种群的稳定性,是提高系统去除ARGs效率的关键[48,50].

在系统设计和操作条件方面,反应器的设计直接影响微藻和细菌的接触效率[23]. 反应器的类型(如鼓泡柱式、平板式、搅拌罐式与气升式等)、流体流动模式以及气体曝气方式等因素都会直接影响微藻和细菌的相互作用效率[55~57]. 合理的反应器设计能够提高微藻与细菌之间的接触面积,从而提升ARGs的去除效果. 在城镇污水处理过程中,微藻-细菌共生系统常采用的反应器结构包括开放式的高速率藻塘(high rate algal pond,HRAP)和封闭式的光生物反应器(photobioreactor,PBR)[23,58]. 以上反应器运行时通常依赖自然光照,以实现经济高效的氧气产生[23]. HRAP一般设计为浅池形式,深度控制在0.2~0.6m范围内,这样有利于阳光的充分穿透,促进藻类的快速生长,并有助于病原体的灭活[59]. 此外,低能耗和慢速旋转的桨轮可实现水体的混合,防止厌氧区的分层和形成. Ovis-Sánchez等[60]研究采用了两套中试规模的室外HRAP系统以平行样方式运行,结果表明HRAP系统可稳定降低总ARGs丰度,进水与出水的ARGs含量对比显示其去除倍数最高达5倍. 相比开放式系统,PBR为藻类和细菌提供了更加稳定可控的生长环境(如光照、养分和温度等)[23]. 单藻PBR对ARGs展现出更优的去除性能,其ARGs去除可降低0.5~3个数量级,而常规活性污泥法污水处理厂往往导致某些ARGs丰度增加或去除效率较低,显著低于单藻PBR的处理效果[61]. 平板式PBR是常用于废水处理和微生物培养系统的反应器形式,其设计以平板状构件为核心,通常呈垂直或倾斜布置,以增加比表面积,优化光照条件、气体交换和污染物接触效率[62,63]. 与其他反应器类型(如序批式反应器或气升式反应器)相比,平板式PBR结构简单和易于规模化,并且在传质效率上具有优势. 然而,在需要高湍流条件的情况下,其混合性能可能不及气升式系统[64]. 除了反应器构型,操作条件也起着至关重要的作用,包括反应器的运行时间、进水流量和氧气供应等,以上因素直接决定微藻和细菌的代谢活动效率[65]. 适当的水流速率和气体曝气能够提高系统的混合性,增加微藻与细菌的接触面积,从而提高ARGs的去除效率.

与此同时,ARGs的类型和复杂性是影响去除效果的重要因素. 不同类型的ARGs具有不同的化学结构、功能和降解难度[66],某些ARGs可能比其他类型的抗性基因更难以降解,这直接影响菌藻共生系统的去除效果. 某些耐药性基因可能需要更长时间或复杂的降解路径才能去除,因此ARGs的种类和浓度会影响系统的降解效率[67,68]. 多重耐药性细菌和ARGs的存在会增加系统的处理难度,因为多重耐药性基因通常比单一的耐药性基因更难去除. 为应对这一挑战,菌藻共生系统可能需要更多种类的微藻和细菌进行协同作用,以提升去除效果. 菌藻共生系统在消减耐药基因方面的效果受多重因素的影响. 通过优化环境条件、选择合适的菌种、改进系统设计以及针对不同类型的ARGs调整处理策略,可以有效提高系统的去除效率.

1.4 微藻及细菌种类的选择及工程应用

菌藻共生系统去除ARGs的效果与微藻与细菌的种类密切相关. 在废水处理及ARGs去除等场景下,优先选择具备高耐逆性和稳定代谢能力的微藻种类,典型代表包括小球藻属(Chlorella)、栅藻属(Scenedesmus)以及链带藻属(Desmodesmus)[69~73]. 这类微藻不仅能耐受污水中高浓度有机物和抗生素残留等胁迫环境,还可通过光合作用持续产氧以支撑细菌有氧代谢,同时释放碳水化合物和氨基酸等有机物质为细菌提供碳源,更能生成活性氧(reactive oxygen species,ROS)抑制携带ARGs的细菌生长并促进游离ARGs降解[74],为系统功能奠定基础. 针对细菌而言,则优先选择兼具ARGs降解能力、营养循环功能及种间协同作用的类群,主要包括芽孢杆菌属(Bacillus)和假单胞菌属(Pseudomonas)等[48,70,72]. 以上细菌通过功能互补与微藻形成代谢协同促进ARGs的去除. 普通小球藻(Chlorella vulgaris)与地衣芽孢杆菌(Bacillus licheniformis)构建的共生体是ARGs去除领域具有代表性的组合,已有研究明确证实其在ARGs去除中的有效作用. 例如,Tang等[48]发现其可高效去除富营养化水体中优势磺胺类抗性基因sul1,对携带sul1的外源ARGs质粒去除率达97%且能抑制sul1复制. Gan等[75]将其应用于城市水体,也观察到多数典型ARGs与移动遗传元件被去除和潜在ARGs宿主相对丰度降低1~4个数量级,仅macB等少数ARGs因土著微生物影响升高. 自然条件下,水体中的天然菌藻共生体群落结构易受环境波动干扰,功能微生物占比低且代谢协同性弱,难以形成稳定高效的共生系统,无法满足污水中ARGs等污染物去除的工程需求. 因此需通过人工干预构建功能明确的工程化共生体系. 工程应用中常优先筛选并引入环境适应性强和协同效应显著的特定藻菌组合,例如小球藻-芽孢杆菌和栅藻-芽孢杆菌等实验室验证类群[76~79],通过外源接种或定向驯化突破天然体系启动缓慢和功能不稳定的瓶颈. 相较于单纯的藻菌混合体系,微藻与活性污泥的组合展现出更优的工程应用潜力,其中以藻菌颗粒污泥的应用优势尤为突出.

2 菌藻共生系统消减抗性基因的机制

菌藻共生系统通过微藻与细菌之间的协同作用,在消减ARGs方面展现了显著的潜力. 如图 3所示,微藻和细菌的共生关系是复杂且互补的,二者通过物质交换、代谢合作以及生态调控等多种机制,协同作用来降解和捕获ARGs,从而有效减少环境中抗性基因的扩散. 微藻通过光合作用将光能转化为化学能,释放氧气和有机碳. 微藻在光合作用过程中生成的氧气不仅能够为细菌提供呼吸所需的氧气,促进细菌的代谢,还可以通过增强细菌的有氧代谢作用,进一步提高细菌对有机物和污染物的降解能力[54]. 微藻产生的有机碳则为细菌提供了代谢的能源,支持其降解环境中的有机污染物,尤其是抗生素及其衍生物[80]. 在这种互利共生关系中,细菌不仅为微藻提供二氧化碳,也促进了微藻的生长. 因此,微藻和细菌之间的营养物质交换是微藻-细菌共生系统去除ARGs的基础,确保了系统的稳定性和高效性. 此外,微藻在其代谢过程中还会产生活性氧(reactive oxygen species,ROS)[81,82]. 微藻产生的ROS可通过氧化损伤ARGs及其携带菌的细胞结构,诱导细胞内氧化应激反应,破坏DNA,尤其是ARGs所在的遗传片段,从而抑制其表达并削弱细菌的增殖能力. 此外,ROS还能破坏细胞膜结构并干扰生物膜的形成,间接降低耐药菌的生存与传播潜力,减少ARGs在环境中的累积[82,83]. 除了通过直接的化学反应破坏ARGs和抗性菌,微藻还通过协同降解抗生素[71,84],降低环境中抗生素的浓度,从而减少ARGs的选择压力[71,85]. 抗生素在水体中的浓度对耐药菌的选择和扩展起到了促进作用,因此,通过降解抗生素,微藻能够减少抗性基因的选择压力,抑制耐药菌的繁殖和基因的扩散. 此外,微藻还可以通过吸附作用帮助去除水中的抗生素,降低其对抗性细菌的滋生作用. 微藻和细菌在共生系统中的互动不仅局限于营养物质的交换,它们之间还通过QS等信号分子调控对方的代谢和生长. 微藻分泌的代谢产物,如某些多糖和环肽,能够调节细菌的QS系统,抑制细菌的生长和抑制质粒的转移作用,进而间接抑制ARGs的水平转移[48,86]. 微藻-细菌共生系统还通过改变微生物群落结构来改变ARGs的水平转移[87]. 菌藻共生系统通过胞外聚合物以及其它分泌物的作用,增强了对ARGs载体(如质粒)的吸附能力[88,89]. 微藻和细菌通过共生体的结构增加了对携带ARGs的外源质粒的吸附作用,从而限制了质粒的转移[90]. 质粒携带的ARGs可以通过跨膜运输进入细菌或微藻细胞[91]. 然而,由于微藻和细菌本身存在多重保护机制,如限制修饰系统、启动子不兼容或转录调控障碍等,这些外源基因在转移后可能难以正常表达,从而降低其活性和传播潜力[92,93]. 微藻-细菌共生系统不仅通过对游离ARGs的物理吸附和化学降解减少其环境残留,还通过生态调控机制限制其在微生物群落中的扩散,有效遏制耐药性基因在环境中的传播风险. 尽管已有的研究揭示了菌藻共生系统在ARGs去除中的多重机制,目前对于菌藻共生系统中跨界胞间通讯作用对ARGs的赋存及传播的影响及其具体机制的理解仍处于初步阶段. 进一步的研究有助于深化人们对这一重要调控机制的认识,并为未来相关防控策略的优化提供理论基础.

3 微藻-细菌跨界胞间通讯作用对耐药基因的影响及机制

微藻-细菌共生体系中的种间通讯对ARGs的表达和传播具有重要影响[50,94]. 细菌通过QS信号分子(如AHLs)协同调控基因表达、质粒转移和生物膜形成,从而影响抗生素耐性和ARGs水平转移[94,95]. 与此同时,微藻分泌的化感物质可以直接或间接干扰细菌QS通路,调节ARGs的传播和消减[13,96]. 图 4中展示了微藻与细菌之间的互利共生不仅促进了彼此的生长代谢,还可能改变细菌QS网络,进而影响耐药基因的动态变化.

细菌的QS系统通过分泌并响应信号分子,感知种群密度,协调包括基因表达、质粒转移和生物膜形成等群体行为. Maseda等[97]和Alcalde-Rico等[98]研究表明,QS系统可显著增强ARGs的表达和传播能力,例如C4-HSL可以刺激MexAB-OprM多药外排泵表达,提升抗生素耐受性. QS还通过调节质粒稳定性和促进共轭转移等机制,加速ARGs的水平传播[99]. QS不仅独立调控HGT行为,还与双组分系统(如QseC/QseB)和信号分子(如c-di-GMP)等网络交互,形成多层级调控[100,101]. Shen等[102]提出在EHEC O157∶H7中,自诱导剂-3激活QseC/QseB双组分系统,上调Ⅲ型分泌系统效应物,增强宿主细胞侵袭和质粒转移效率. 革兰氏阴性菌中,群体感应直接调节Ⅳ型分泌系统活性,控制ARGs的水平转移[103]. 铜绿假单胞菌利用LasI/LasR群体感应系统调节Ⅵ型分泌系统活性,促进胞外囊泡介导的ARGs传递[104]. 以上调控不仅限于种内,在QS信号跨物种识别下,异种间ARGs转移频率也显著上升. Zheng等[105]报道N-辛酰基-L-高丝氨酸内酯(C8-HSL)在100 ng·L-1时可使大肠杆菌属间的接合频率增加28倍. 生物膜由QS系统精细协调形成,是ARGs的重要“庇护所”. 通过限制外部刺激的进入,提升ARGs的持久性和迁移性. QS控制生物膜中细胞间黏附、基质合成与代谢协同,使ARGs难以彻底去除. 同时,生物膜内HGT效率远高于游离状态,加剧环境中耐药基因的传播风险. 因此,针对QS的干预,如生物炭降解C4-HSL等策略,正被探索用于削弱QS主导的ARGs维持与转移[106].

微藻光合作用产生的有机酸和氨基酸等代谢物不仅为细菌提供营养,还可作为QS信号的激活剂或抑制剂,改变细菌群体感应和耐药基因表达. Mazumder等[107]研究发现,微藻释出的有机酸可激活细菌QS受体,从而调节抗性基因的启动转录. QS信号可以影响微藻的生命活动如AHLs在藻菌界面中通过调控杀藻化合物的产生间接影响微藻的生长和代谢[108]. 微藻与细菌协同作用时,信号分子传递可改变细菌外排泵活性,间接抑制ARGs传播,协同去除抗生素并削弱耐药. 微藻可以释放卤代呋喃酮、类胡萝卜素、藻胆蛋白和二十碳五烯酸等生物活性化合物,干扰细菌QS信号,抑制生物膜形成和ARGs表达与转移[96]. Liu等[50]研究表明,与纯细菌体系相比,藻菌颗粒污泥系对四环素和磺胺二嗪相关ARGs和移动遗传元件的相对丰度分别降低了约56.1%和22.1%. 这表明微藻释放的化感物质能够通过调控细菌群体感应信号间接减少耐药基因的传播. 总体来看,微藻通过直接影响细菌代谢和生长,以及间接调控QS网络,显著降低了环境中ARGs的丰度和传播风险[95]. 此外,微藻提供的微环境(如光合作用产生的氧气和代谢产物)可调节细菌的生物膜形成和细胞间接触行为,进一步影响ARGs的水平转移.

群体感应抑制剂(quorum sensing inhibitor,QSIs)通过特异性阻断信号分子的合成、释放或受体结合,切断细菌间的通信,从而抑制群体行为并减少ARGs和VFGs的水平转移. 与传统抗生素不同,QSIs具有“非杀灭性”特点,不会对细菌生长造成直接杀伤,因此不会产生显著的耐药选择压力[109]. 实验表明,包括香兰素、4-硝基吡啶-N-氧化物和AHLs裂解酶等在内的多种QSIs可显著降低菌间和菌内的质粒接合频率. 例如,Qiu等[94]实验得到香兰素在亚抑菌浓度下通过抑制QS信号释放或受体结合,抑制了细菌生物膜形成和外泌毒力因子的合成,从而减少了抗药性基因(如RP4质粒)的转移. QSIs策略为抗耐药基因传播提供了一种新型的生态友好途径,通过干扰群体感应,弱化了ARGs扩散的动力学基础. 微藻-细菌共生体系因其“协同增效”特性而在抗生素去除和ARGs抑制方面展现出巨大潜力. 藻菌颗粒污泥体系在抗生素污水处理和ARGs减量方面效果显著,是一种极具前景的生物处理技术[50,110]. 未来需进一步阐明微藻代谢产物与细菌群体感应信号之间的交互机制,以指导耐药基因防控策略的优化. 同时,探索QSIs在藻菌系统中的应用,尤其是在抑制ARGs传播的同时避免产生新的抗性选择压力,也是研究重点. 通过深入的机制研究和技术创新,微藻-细菌共生系统有望成为一种生态友好和可持续的耐药基因治理手段,为减缓环境中ARGs的扩散提供新思路.

4 微藻-细菌共生系统在ARGs治理中的挑战与发展路径 4.1 微藻-细菌共生系统在ARGs治理中的工程应用与系统优化挑战

微藻-细菌共生系统因其协同代谢与生态调控机制,在污染水体中ARGs的去除方面展现出巨大应用潜力[111]. 然而,将该系统从实验室条件推广至实际工程应用仍面临严峻挑战. 微藻与细菌在共生过程中对环境变化高度敏感,温度、光照和pH等因子的波动可能破坏微生物群落的稳定性,进而影响ARGs的去除效率. 在复杂水质条件下实现系统的长期稳定运行和高效治理能力,是实现工程化应用的关键. 为提升系统适应性与抗干扰能力,模块化设计和智能反馈调节机制已成为关键优化策略. 模块化结构可将系统划分为多个功能单元,便于根据污染物类型和水质条件灵活配置,提升系统的资源利用效率和运行稳定性[112]. 在发生外部扰动或单元故障时,模块化也有助于快速识别问题并进行响应修复[113]. 同时,引入实时传感器与自动控制系统,可对温度、pH和溶解氧等关键参数进行持续监测,并根据污染物浓度变化进行自适应调节,从而保障系统始终处于最优运行状态,提升ARGs的动态去除能力[113]. 除了运行结构与过程控制的优化,系统本身的生物稳定性与功能特异性亦是工程化成功的基础. 为此,构建高稳定性的合成微生物共生体系成为另一核心方向. 通过基因工程手段对共生微生物进行定向改造,例如增强微藻在低光或低温环境下的光合作用效率,或提升细菌对特定污染物的降解能力,可显著增强系统在多变环境中的适应性与代谢协同效率[114,115]. 以上策略有助于构建兼具功能专一性与环境普适性的微藻-细菌共生平台,为ARGs的高效去除与工程化应用奠定坚实基础.

4.2 微生物群落与ARGs传播风险的控制

微藻-细菌共生系统中的微生物群落,不仅能有效去除水中的ARGs,但也可能为ARGs的传播提供温床[116]. HGT是ARGs传播的主要途径,而在微藻-细菌共生系统中,由于其内的微生物种类多样、环境因素以及抗生素在藻层的生物累积的影响,可能会加剧ARGs的传播[83]. 控制ARGs的传播风险是微藻-细菌共生系统应用中的一个重要问题. 为此,需要借助组学技术对微藻-细菌共生系统中ARGs传播的生态机制进行深入研究. 宏基因组学和转录组学等技术可以帮助揭示ARGs传播的具体机制及其受环境因素和微生物相互作用等的影响[117,118]. 通过分析ARGs在微生物群落中的分布和迁移路径,研究人员能够识别出传播的关键因素,进而为系统优化提供理论依据[119]. 同时,开发具有特异性抑制作用的调控因子,对于抑制HGT至关重要. 例如,利用信号分子阻断剂干扰微生物之间的基因交流,能够有效减少耐药基因的传播[120]. 通过这种方式,可以在源头上防止ARGs的水平转移,确保微生物群落的稳定性. 此外,通过精确调整微生物群落的环境条件,特别是在营养成分和pH值等方面的优化,能够抑制耐药菌的生长竞争力,减少其在系统中的扩展空间. 以上技术不仅能够抑制ARGs在微藻-细菌共生系统中的传播,还可以在更广泛的水体治理过程中,减少耐药基因的积累和传播风险,从而确保系统的长期稳定运行.

4.3 微藻-细菌共生系统的成本效益与政策支持

微藻-细菌共生系统相比传统污水处理工艺具有显著的费用优势,主要得益于其光合作用供氧机制. 传统好氧工艺(如活性污泥法)中,曝气供氧通常占总能耗的40%~60%,有时甚至高达50%~90%[121,122]. 微藻-细菌共生系统通过太阳能驱动光合作用供氧,大幅降低了电力需求,提供了零成本的氧化模式[123]. HRAP系统的能耗被报道为0.06~0.25 kW·h·m-3,显著低于传统活性污泥法的0.89 kW·h·m-3[124,125]. Kohlheb等[126]在西班牙一项示范性HRAP工艺(处理能力为300 m3·d-1)通过机械混合优化,能耗降至0.10~0.17 kW·h·m-3,远低于传统工艺的0.3~0.6 kW·h·m-3. HRAP的处理成本约为1.50元·m-3,低于序批式活性污泥法的2.17元·m-3[124]. 另有研究发现,常规活性污泥法的平均处理成本为6.60元·m-3,而HRAP为3.51元·m-3,仅为前者的一半[126]. 突尼斯大学合并氧化HRAP示范装置(含鼓风设备)的能耗为1.05 kW·h·m-3,运行成本为2.42元·m-3,规模优化后有望进一步降低[127]. 微藻-细菌共生系统的主要构成是浅塘,土建和设备设计较为简单,单位容积的投资低于机械化工艺.

尽管微藻-细菌共生系统在ARGs治理以及污水处理成本方面展现出显著潜力,但其在实际应用中的推广仍面临较高的初期建设和运营成本,以及经济与政策层面的多重障碍[128]. 其初期投资主要集中在设备建设、研发投入和系统安装与调试等环节,且由于技术尚未完全成熟,缺乏统一的操作规范,也进一步抬高了运行维护成本. 生物质收获环节约占系统总运行成本的20%~30%,是限制其经济可行性的关键瓶颈之一[113]. 然而,通过降低曝气能耗和减少CO2排放,并探索其产生的生物质用于高值化资源回收(如生物燃料生产)的潜力,该系统仍具备显著的成本优化前景[128]. 为克服上述经济与政策障碍,政府与产业界的协同合作至关重要[129]. 通过建立示范项目,可验证系统的技术可行性与经济效益,并为政策制定提供实践依据. 政府可通过资金扶持和税收减免等激励措施,降低企业投资风险,吸引更多社会资本参与技术推广. 同时,制定统一的技术标准与行业规范,有助于规范市场运行和推动技术健康发展. 将微藻-细菌系统与现有污水处理工艺(如膜生物反应器)结合,构建低成本和高效率的商业模式,是提升市场接受度的有效路径[130]. 例如,结合高效率藻塘与膜生物反应器的系统可实现高达90%的化学需氧量(COD)去除率,并有效降低运行成本[113,131].

4.4 未来研究方向与技术融合趋势

如图 5所示,未来研究应聚焦于以下方向以提升微藻-细菌共生系统在ARGs治理中的应用效果. 首先,应深入解析ARGs去除的核心机制,特别是微藻与细菌间的代谢互作及其对系统协同效能的影响[85]. 其次,融合膜反应器和电化学技术等先进污水处理工艺,有望显著提升系统的处理能力与适应性. 此外,应优先探索细胞间通讯调控机制在菌藻系统中的整合路径,特别是在干扰QS信号方面. 例如,结合CRISPR-Cas等基因编辑工具与高通量筛选技术,可用于定向优化共生菌株,增强其对QS信号的识别与抑制能力,从而提高ARGs的去除效率[132]. 人工智能与机器学习技术的引入也为系统运行的智能优化提供了新机遇[133,134]. 通过实时数据分析与反馈机制,AI可动态调节操作参数,优化系统状态. 例如,基于环境变量(如营养比例、光照强度和温度等)的预测模型可辅助识别ARGs扩散风险,进而实施自适应干预[135,136]. 为实现该技术的广泛应用,尤其是在资源有限的发展中国家,推动微藻-细菌共生系统的本地化低成本部署也至关重要. 通过技术简化、材料替代与商业模式创新,可提升其区域适应性与经济可行性,最终实现在全球范围内对ARGs污染的高效治理.

5 结论

本文综述了微藻-细菌共生系统在ARGs治理中的应用潜力,揭示其通过代谢协同作用与细胞间信号调控机制实现ARGs有效去除的多重路径,为污水处理提供了绿色和可持续的新思路. 菌藻共生系统依托微藻与细菌之间的功能互补与信号调节,在降低ARGs浓度和抑制其水平转移方面展现出显著优势. 其中,QS系统被认为是影响ARGs传播与持久性的关键调控机制. 微藻可通过分泌QS信号分子类似物或调节代谢产物干扰细菌QS过程,从而抑制生物膜形成和减少质粒转移. QSIs的引入进一步拓展了该系统在ARGs防控中的应用策略,也为其优化提供了技术支撑. 尽管如此,微藻-细菌共生系统在实际应用中仍面临诸多挑战,如系统稳定性受环境扰动影响显著、规模化运行难度大以及运行成本偏高等问题. 为解决以上瓶颈,未来应加强模块化设计和构建高稳定性的合成共生体系,并引入实时监测与反馈调节机制,提升系统的适应性与鲁棒性. 此外,深入探索微藻-细菌之间的信号传导机制,特别是微藻在QS调控中的作用,以及QS抑制剂的高效筛选与定向利用,将为ARGs治理提供更加精准的调控工具. 通过多学科技术融合与系统优化,微藻-细菌共生系统有望成为全球ARGs污染控制与水体生态安全保护中的重要解决方案.
