全氟化合物(perfluorinated compounds,PFCs)是一类难以降解的持久性有机污染物,其在全球范围的水体[1]、 大气[2]、 土壤[3]、 生物体[4]等各种基质中均有一定的检出. 由于具有优良的热稳定性和表面活性,PFCs广泛地应用于纺织、 皮革、 造纸、 电镀等行业的生产中[5, 6]. PFCs可伴随生产过程和产品使用过程释放到环境中. 与全氟羧酸(perfluorinated carboxylic acids,PFCAs)和全氟磺酸盐(perfluorinated sulfonates,PFSAs)化合物相比,全氟磺酰胺类和全氟调聚醇类因具有低水溶性和高挥发性而更易进入大气中,通过大气光化学反应及干、 湿沉降进入地表水、 土壤等环境基质进一步发生各种化学反应降解成较为稳定的PFCAs和PFSAs[7, 8, 9, 10],因此,PFCAs和PFSAs往往是除大气以外其他环境基质中PFCs污染调查时研究的重点.
作为PFCs前体,全氟辛基磺酰氟(perfluorooctyl sulfonyl fluoride,POSF)的生产已有40多年的历史. 其广泛地应用于电化学氟化生产中. 到2002年为止,估计约有96000 t POSF用于生产中[11]. 由于其代谢的PFCs具有很强的生物累积性[12],自2002年起3M公司已逐步停止POSF的生产,但其他相关的PFCs仍在不断生产[13]. 近10年来,PFSF的生产已由北美和欧盟转向我国. 2004年至2006年,我国POSF的产量增加了4倍[14],当时每年的产量超过200 t[15],其中约100 t出口到南美以及部分欧洲地区[15]. 因此,研究我国PFCs的环境污染问题是十分必要的.
目前,关于水体PFCs污染的研究主要围绕着经济发达地区的内陆河流、 河口及城市污水处理厂排放的废水[16, 17],关于海洋中PFCs的来源、 分布和环境行为的报道较少[18]. 现有的关于我国海洋环境PFCs的污染的报道主要集中在香港周边海域[19]、 大连湾[20]、 东南沿海[14]和北渤海[21],但这些报道都基于表层海水中PFCs的研究,并未对海洋沉积物PFCs的污染状况和PFCs在海水-沉积物上的分配进行深入探讨. 深圳地处广东省南部,为华南地区的第二大城市,毗邻香港. 其近岸海域PFCs污染状况近年来鲜有报道. 本研究以深圳近岸海域大亚湾、 大鹏湾、 深圳湾和伶仃洋为研究海域,分别采集10份表层海水和7份表层沉积物,分析PFCs的含量,探讨其污染特征、 来源及在水-固相间的分配,以期为我国PFCs的污染状况和控制提供重要参考数据.
1 材料与方法 1.1 试剂与标样
本研究实验所需标样购自加拿大Wellington公司,包括16种PFCs外标和8种PFCs内标,具体见表 1. 甲醇(色谱纯)购自美国J. T. Baker公司,甲酸(96%,色谱纯)和醋酸铵(色谱纯)购自美国Dikma公司,氨水(优级纯)购自比利时Acros公司,Qasis WAX固相萃取柱(6 cm3,150 mg)购自美国Waters公司,实验用水为电阻率18.2 MΩ ·cm的超纯水.
2013年9月乘船在深圳距岸2 km以外近岸分别采集10份表层海水(0~20 cm)和7份表层沉积物(0~10 cm). 采样站位距岸2 km以上,具体采样点见图 1. 每份表层水样以不锈钢桶进行采集,采样量约为2 L,采集后的水样装入聚丙烯瓶中. 每份表层沉积物样品以不锈钢抓斗采泥器进行采集,每份样品采样量约为1 kg,采集后装入聚丙烯密实袋中. 所有的采样工具和承装袋预先用甲醇和超纯水清洗,装样前再用海水清洗. 水样运回实验室后在4℃下避光保存,沉积物样品冷冻干燥后,研磨,过0.15 mm孔径筛,装入聚丙烯袋于-20℃保存,待分析.

水样和沉积物中PFCs的提取方法分别参见文献[22, 23]. 简言之,取1 L水样经0.45 μm孔径滤膜进行过滤,滤后水样以2% 甲酸溶液调节pH至3,加入5 ng内标物,上固相萃取柱,先后以2%甲酸溶液、 2%甲酸甲醇溶液(1 ∶1)、 甲醇溶液淋洗,后用9%氨水甲醇溶液洗脱,用氮气吹干后以甲醇定容至1 mL,待测. 取1 g混匀沉积物样品于离心管中,加5 ng内标和10 mL甲醇,振荡提取,超声萃取并离心,重复提取两次后合并上清液于50 mL离心管中,以氮气吹干并加入8 mL 2%甲酸溶液和42 mL超纯水,上固相萃取柱进行萃取,萃取方式和后续步骤与水样PFCs提取方式相同. 1.4 仪器分析
PFCs分析测定采用液相色谱质谱联用仪(HPLC-MS/MS)测定,液相色谱为Agilent technologies 1290,质谱为 QTRAP 5500三重四级杆串联质谱系统(美国AB公司),色谱柱为 Agilent poroshell 120 EC-C18(100×2.1 mm,2.7 μm). HPLC系统的修正方法见文献[24]以消除仪器空白的干扰. 流动相A选择5 mmol ·L-1醋酸铵水溶液,流动相B选择5 mmol ·L-1醋酸铵甲醇溶液,流速为0.2 mL ·min-1,进样体积10 μL. 采用二元梯度洗脱方式,洗脱程序为:0~3 min,流动相A由90%降至30%,流动相B由10%升至70%; 3~13 min,流动相A由30%降至0%,流动相B由70%升至100%; 13~14 min,流动相A由0%升至90%,流动相B由100%降至10%; 14~20 min,流动相A保持在90%,流动相B保持在10%. 质谱条件为:采用电喷雾离子源(ESI),负离子扫描方式,多反应监测模式(MRM),离子源温度为500℃,离子喷雾电压为-4500 V,气帘压力为276 kPa,雾化气压力为414 kPa,去溶剂压力为276 kPa,内标法定量,质谱定量母离子/子离子(m/z)对见表 1,其他质谱参数见文献[25]. 1.5 质量保证与质量控制
为保证实验结果的准确性,本实验所用实验器皿均选择聚丙烯材质,使用前用甲醇和超纯水淋洗. 为避免样品提取过程中的外源性污染,每批样品(3~4个)进行全程空白实验,实验基质为超纯水和以甲醇淋洗后的表层沉积物. 所有实验空白基质中PFCs含量均低于仪器检测限(instrument detection limits,IDLs)或未检出(not detected,n.d.). 采用基质工作曲线-内标法定量,分别以3倍和10倍信噪比(signal-to-noise ratio,S/N)确定目标分析物的IDLs和方法定量限(method quantification limits,MQLs). 16种PFCs在水样和沉积物中的IDLs和MQLs及加标回收率列于表 2.
1.6 统计分析
数据采用IBM SPSS 20.0(美国IBM公司)和Origin 9.0(美国Origin公司)软件进行统计分析. 统计分析时,若PFCs含量低于IDLs或未检出时,其含量数值以0表示; 若PFCs含量高于IDLs而低于MQLs,其数值以1/2 MQLs表示[26].
2 结果与讨论 2.1 表层海水中PFCs的质量浓度
表层海水中检测了16种PFCs,有10种不同程度地检出,分别为PFPeA、 PFHxA、 PFHpA、 PFOA、 PFNA、 PFDA、 PFUdA、 PFBS、 PFHxS和PFOS. 其中,PFOA、 PFNA、 PFBS、 PFHxS和PFOS 100%检出. 检出的10种PFCs的质量浓度统计结果及其组成与空间分布分别示于图 2(a)和图 3(a). 深圳近岸海域表层海水ΣPFCs质量浓度范围在1.74 (O10)~14.7 (O5) ng ·L-1之间. 伶仃洋、 深圳湾ΣPFCs质量浓度显著高于大亚湾、 大鹏湾(P<0.05),呈西高东低的分布态势,且距岸越远ΣPFCs质量浓度越低[图 3(a)]. 这与河流入海的PFCs输入有关. 西部海域的入海河流主要为珠江和东宝河,由于它们主要流经广州、 东莞和深圳,这些城市排放的工业废水和生活污水使河流本身具有高浓度的PFCs[27],这些河流入海后导致伶仃洋、 深圳湾表层海水PFCs质量浓度较高. 而汇入大亚湾的河流主要为流经惠州的淡水河,惠州为旅游型城市,大亚湾也建成了风景区,这些区域受工业排污影响较小,因而其近海域PFCs污染程度也较低. 受洋流的稀释以及吸附、 降解和生物富集作用,进入海洋的PFCs质量浓度随离岸距离的增加逐渐降低.

PFBS、 PFOS和PFOA是表层海水3种主要的PFCs,平均质量浓度分别为2.57、 1.32和1.30 ng ·L-1. 在中国的部分河流和海洋PFCs质量浓度研究中,PFOA和PFOS均为主要PFCs[22, 26, 27, 28, 29, 30],但近年来我国部分内陆水体[31]、 海洋[14]PFCs质量浓度研究表明,PFBS替代PFOS成为入海口上游河流和表层海水中主要的PFSA,本研究也与之相符. 这主要与N-甲基全氟丁基磺基乙醇胺(N-methyl perfluorobutane sulfonamidoethanol,MeFBSE)及其他相关的全氟四碳化合物的增产有关[14],这些产品是在2002年3M公司逐步淘汰生产POSF后引入的[32]. PFOA(0.296~2.05 ng ·L-1)是表层海水质量浓度最高的PFCA,其次为PFNA(0.411~0.697 ng ·L-1)、 PFPeA(<0.0397~0.932 ng ·L-1)、 PFUdA(n.d.~0.467 ng ·L-1)、 PFHxA(<0.0266~0.901 ng ·L-1)、 PFUdA(n.d.~0.467 ng ·L-1)和PFHpA(<0.0182~0.337 ng ·L-1),12碳链以上的PFCAs未检出.
从PFCs组成上看,表层海水中PFBS所占比例最高,为20.9%~46.9%(均值为32.5%),其次为PFOS(12.0%~35.5%,均值为21.9%)、 PFOA(14.7%~21.7%,均值为19.0%)、 PFNA(4.36%~24.0%,均值为11.8%). 其他PFCs单体所占比例较低,平均质量浓度所占ΣPFCs比例小于5%. 2.2 表层沉积物中PFCs的含量
表层沉积物中检测的16种PFCs,有8种不同程度检出,包括PFPeA、 PFHxA、 PFOA、 PFNA、 PFDA、 PFUdA、 PFHxS和PFOS. 除PFUdA外,其他PFCs 100%检出. 8种检出的PFCs含量统计结果及其组成与空间分布见图 2(b)和3(b). 与表层海水ΣPFCs的质量浓度分布不均不同,表层沉积物ΣPFCs含量范围在2.22 (O6)~2.62 (O2) μg ·kg-1之间,其变化范围很小(变异系数为6.34%). 由于调查海域距陆地有一定的距离(>2km),来自于近海工业废水PFCs的排放对海洋沉积物的影响较小,因而本次调查采集沉积物中的PFCs主要来自于对上覆水PFCs的吸附以及吸附了水中PFCs的颗粒物的沉积. Zhao等[33]研究了不同比重和粒度的沉积物对PFCs的吸附. 结果表明,疏水作用在沉积物吸附长链PFCs过程中起支配作用,而静电引力是控制短链PFCs吸附于沉积物的主要因素,且沉积物吸附PFSAs的量远高于同碳链长度的PFCAs. 本次调查的海洋表层沉积物TOC质量分数变化非常小(0.831%~0.923%),由图 2(b)可知,长链PFCs含量变化非常小,检出的PFNA、 PFDA和PFUdA变异系数分别为3.21%、 10.7%和4.57%. 而短链的PFCs由于受静电引力作用,其在沉积物中含量变化较长链大,PFPeA、 PFHxA和PFOA变异系数分别为15.9%、 33.9%和51.8%. 而对于PFSAs,由于沉积物对其吸附能力远高于PFCAs,因而PFHxS和PFOS含量变化也不大,变异系数分别为1.35%和11.7%. 总体来说,由于受陆源影响较小,沉积物对上覆水PFCs的吸附和解吸作用也处于动态平衡,因而各采样点表层沉积物PFCs含量变化不大.
PFOS是表层沉积物中最主要的PFCs,平均含量为0.780 μg ·kg-1. 表层水中高浓度的PFBS在沉积物中未检出,表明极短链的PFCs更容易分配在水相而难于被沉积物吸附. PFNA(0.312~0.337 μg ·kg-1)是含量最高的PFCA,其次为PFDA(0.214~0.275 μg ·kg-1)、 PFUdA(n.d.~0.276 μg ·kg-1)、 PFHxA(0.158~0.350 μg ·kg-1)、 PFOA(0.136~0.451 μg ·kg-1)和PFPeA(0.138~0.212 μg ·kg-1),它们的平均含量相差很小. PFPeA和12碳链以上的PFCAs未有检出.
从PFCs组成上看,表层沉积物中PFOS所占比例最高,为28.0%~38.5%(均值为32.3%),其次为PFNA(12.4%~14.8%,均值为13.5%)、 PFHxS(9.26%~11.0%,均值为10.2%)、 PFDA(8.14%~12.0%,均值为10.1%)、 PFUdA(0~11.6%,均值为9.27%)、 PFHxA(6.12%~13.2%,均值为9.24%)、 PFOA(5.76%~17.4%,均值为8.69%)和PFPeA(5.91%~8.08%,均值为6.74%). 除PFOS外,其他PFCs单体组成上差别不大. 2.3 PFCs在沉积物和液相间的分配系数
以同一采样点沉积物和水相PFCs的含量来计算PFCs的分配系数. 分配系数(Kd)和标准化分配系数(Koc)依下式来计算[34, 35]:

式中,cs为沉积物PFCs的浓度(μg ·kg-1),cw为液相PFCs的浓度(μg ·L-1),foc为沉积物中有机碳的质量分数(%),Kd和Koc的单位为 L ·kg-1.
表层沉积物检出的 PFCs的lgKd和lgKoc值示于表 3. 本研究各PFCs单体的lgKd和lgKoc与海河和大沽运河研究结果相近[36],但远高于其他文献报道的lgKd值和lgKoc值[34, 35, 37]. 不同研究中的PFCs的lgKd值的差异可能与PFCs的物理化学特性有关. 由于PFCs属于阴离子表面活性剂,疏水作用和静电引力同时对其在水环境中的吸附行为存在影响[36],PFCs的碳链的长度也显著地影响其在沉积物上的吸附[38]. 实际环境中沉积物的性质和水环境条件也对PFCs的分配存在显著影响[26],如水的pH、 Ca2+、 盐度和沉积物的有机碳的含量等参数都会显著地影响PFCs在沉积物上的吸附[18, 38, 39].
本研究条件下,PFCAs单体在沉积物上的吸附显著受碳链长度的影响,lgKoc均值从4.35 L ·kg-1(PFOA)增加到4.95 L ·kg-1(PFDA). 除此之外,磺酸基和羧酸基官能团的影响也是至关重要的,具有相同碳链长度的PFSA的lgKoc均值较PFCA高 0.5~0.8,说明PFSA较PFCA更倾向于积累在沉积物中,这与之前的研究结果是一致的[38].
不同海域关于PFCs的污染特征并不一致,如中国东海-南海[14]、 德国湾及波罗的海[40]的表层海水中PFOA、 PFHpA和PFBS为主要PFC单体,而地中海[41]、 韩国[42]、 东京湾[43]和西班牙Cantabrian海[44]的表层海水则以PFOS、 PFOA为主要PFCs. 本研究表层海水主要PFCs为PFBS、 PFOA和PFOS. 不同海域PFCs污染物构成的差异可能与PFCs的区域来源有关. 此外,洋流、 颗粒物的吸附及大气沉降等因素也影响着海洋水体PFCs的组成[14]. 本研究表层海水PFBS的平均质量浓度约为PFOS和PFOA的两倍,这与MePBSE及其他相关的全氟四碳化合物的增产有关[14]. 由于世界许多国家水体中PFOS、 PFOA主要来自于污水处理厂排放的废水[45, 46, 47, 48],因而深圳近岸海域表层海水PFOA和PFOS可能主要来自于污水处理厂排放的废水. 不同采样点表层海水中PFCs的主要单体浓度相关关系见表 4. PFBS、 PFOA、 PFOS和PFNA均呈显著正相关(P<0.01),说明这些化合物具有同源性. Young等[49]在从北极圈取得的雪样中发现PFNA和PFOA呈非常显著正相关,其斜率约为1,此样本中的PFCs仅来自于大气中的全氟调聚醇类化合物的降解. Ahrens等[50, 51]在研究大西洋PFCs来源时发现PFNA和PFOA也呈正相关(斜率约为0.4),进而发现若PFNA和PFOA的相关关系的斜率越接近于1,其来自大气沉降的可能性越大. 本研究PFNA和PFOA相关关系的斜率为0.112,表明深圳近岸表层海水中PFCs存在除大气沉降外更重要的污染源. Simcik等[52]指出高PFHpA/PFOA比值可作为PFCs来自于大气沉降证据,但本研究中仅O1和O5两个点位PFHpA的含量高于MQL,其PFHpA/PFOA分别为0.164(O1)和0.0971(O5),这表明深圳近岸海域表层海水PFCs主要来自于污水排放,大气沉降来源极少.
由于采样点距离海岸距离海岸2 km外,且采样区域并不属于港口,来往船只较少,采样区域也不存在垃圾倾倒,因而来自于陆源固废和河流携带泥沙中PFCs含量可以忽略,海洋表层沉积物中PFCs主要来自于沉积物对上覆水中PFCs的吸附. 2.5 表层海水和表层沉积物中PFCs含量的比较
已报道的全球不同近岸海域表层海水中PFBS、 PFOS、 PFOA质量浓度数据列于表 5,表层沉积物中PFOS、 PFOA含量数据列于表 6. 由表 5可见,不同近岸海域表层海水PFCs特征污染物不同. 日本沿海区域和东京湾的PFOS、 PFOA质量浓度较高,德国湾以PFBS和PFOA为主,中国大连湾以PFOA为主,而深圳近岸海域以PFBS为主. 这反映了不同海域PFCs的污染特征和来源的不同. 由表 6可知,旧金山湾和东京湾表层沉积物PFOS污染较其他区域严重,德国湾和Areake海PFOA含量高于其他调查海域,其他海域表层沉积物PFOA含量相差不大. 总体上,不同近岸海域沉积物中PFOS和PFOA的含量变化远不如表层海水变化大,本研究结果也再次印证了这一点. 说明污染源较远的情况下,近岸海域表层沉积物中的PFCs主要来自于对上覆海水中PFCs的吸附,吸附量的大小主要取决于沉积物的性质、 PFCs单体碳链长度和海水pH、 盐度等参数.
3 结论
(1)深圳近岸海域普遍检出PFCs化合物. 表层海水ΣPFCs质量浓度在1.74~14.7 ng ·L-1之间,PFBS、 PFOS、 PFOA为主要PFCs. 表层沉积物
ΣPFCs含量在2.22~2.62 μg ·kg-1之间,PFOS为主要PFCs.
(2)表层海水中ΣPFCs质量浓度变化较大,深圳湾、 伶仃洋显著高于大亚湾、 大鹏湾(P<0.05); 表层沉积物ΣPFCs含量变化较小.
(3)沉积物吸附PFCs的能力随碳链长度增加而增强,且相同碳链长度的情况下,PFSAs较PFCAs更易于被吸附.
(4)表层海水中PFCs主要来自于污水排放和河流入海的输入; 表层沉积物中PFCs主要来自于其对上覆海水PFCs的吸附.
(5)深圳近岸海域表层海水PFBS污染较为严重,而沉积物PFOS含量与其他海域相差不大.
